Die sich wiederholende Folge der Querstreifung wird als Sarkomer bezeichnet, wobei die Zwischenscheibe (Z-Streifen) als Grenze gilt. Der isotope Streifen (I-Streifen) wird durch den Z-Streifen halbiert. Der anisotope Streifen (A-Streifen) ist elektronendichter und wird durch ein schmales, helles Band, den H-Streifen unterteilt. Entlang der Mittellinie des H-Bandes erscheint häufig ein M-Streifen. Neben den Fibrillen ist eine deutlich sichtbare vesikuläre Komponente im interfibrillären Zytoplasma zu erkennen, das sarkoplasmatische Retikulum, das in Höhe der Z-Linie unterbrochen ist. In dieser Höhe wird das sarkoplasmatische Retikulum durch erweiterte Säckchen, die terminalen Zisternen repräsentiert, die eng einem querverlaufenden T-Membran-System anliegen.

Zwei terminale Zisternen und ein T-Tubulus bilden jeweils eine Triade. Während der Skelettmuskel der Säuger zwei Triaden pro Sakomer besitzt, und diese in Höhe des Übergangs vom A- zum I-Streifen liegen, kommen im hier gezeigten Amphibienmuskel nur eine Triade pro Sakomer vor.

Transmission electron microscope (TEM)

The sections for examination under the transmission electron microscope are about 0.1µm thick. They are referred to as ultra-thin sections.

To obtain such ultra-thin sections, the tissues are embedded in plastic polymers like epon (instead of paraffin, which is used for examination under the light microscope) after fixation and dehydration. Ultra-thin sections are not stained with dyes, but contrasted with heavy-metal salts. The heavy-metal salts lead to a different electron scatter and thereby create a differentiated blackening of the photographic negative. A common method of creating contrast results with 5% uranyl acetate and lead citrate.

Die Kontraktion der Skelettmuskulatur wird über die motorischen Endplatten eingeleitet und führt zu einer Depolarisation der Zellmembran (Sarkolemm). Diese wird über das T-Tubulus-System (transversales oder T-System), in das Zellinnere weitergeleitet. In Höhe des Übergangs vom A- zum I-Streifen liegen die T-Tubuli zwischen den Myofibrillen direkt zwei terminalen Zisternen des sarkoplasmatischen Retikulums an und bilden mit diesen eine Triade. Diese Triaden kontrollieren die Muskelkontraktion, indem sie die Calciumfreisetzung regulieren. Sie bewirken die elektromechanische Koppelung, d. h. die gleichzeitige Kontraktion aller Myofibrillen eines Faserquerschnitts.
