Die längs geschnittenen glatten Muskelzellen des Stratum circulare lassen deutlich die fein strukturierten stäbchenförmigen Zellkerne erkennen. Die Enden sind zum Teil geschlängelt oder korkenzieherartig gewunden. Das Zytoplasma der glatten Muskelzellen erscheint homogen und läßt keine Querstreifung erkennen. Jede der angeschnittenen polygonalen Zellen des Stratum longitudinale ist von feinem, blau gefärbten Bindegewebsfasern umgeben.

Cell nuclei are stained with hemalaun or Weigerts iron-hematoxylin and then counterstained with a dye mixture of orange-phosphormolybden with aniline blue.

Der glatten Muskelzelle fehlt im Vergleich zur Skelettmuskulatur die strenge Anordnung der kontraktilen Elemente, die aus Actin- und Myosinfilamenten bestehen. Diese verlaufen zusammen mit den Intermediärfilamenten kreuz und quer durch das Zytoplasma, bilden ein Netzwerk, dessen fokale Verdichtungen sich an der Zellperipherie verankern. Über die fokalen Verdichtungen werden bei der Kontraktion die Spannungen auf die die Muskelzellen umgebenden Laminae externae weitergegeben. So wirken viele glatte Muskelzellen als Funktionseinheit.
