Durch die unterschiedliche Anfärbbarkeit von kollagenen Bindegewebsfasern und Muskulatur bei der Trichromfärbung nach van Gieson sind diese beiden Gewebebestandteile leicht voneinander zu differenzieren. Dies wird am Beispiel der Zunge gezeigt. Unter dem verhornten mehrschichtigen Epithel der fadenförmigen Zungenpapillen des Zungenrückens befindet sich eine Bindegewebsschicht. Diese enthält dicht gepackte kollagene Fasern, die sich intensiv rot anfärben. Darunter folgt die quergestreifte Muskulatur des Zungenkörpers, deren einzelne Züge in Längs-, Quer- und Vertikalrichtung angeordnet sind. Die Muskulatur färbt sich gelb an, das Bindegewebe rot, die Epithelzellen der Zungenschleimhaut und ihre Zellkerne sind gelb- bis schwarzbraun.

This is the dye most commonly used to stain connective tissue.

Dyes:

Weigerts iron hematoxylin This dye is one of the hematoxylins, but stains much more intensively than hemalaun. It is used with van Gieson’s dye to stain cell nuclei.
Picro-fuchsin or Picro-thiacid mixture This dye solution simultaneously stains connective tissue red and muscle tissue yellow (simultaneous stain).
Die fadenförmigen Zungenpapillen (Papillae filifomes) sind die kleinsten, aber bei weitem zahlreichsten der vier Papillentypen der Zunge. Das verhornte Epithel der fadenförmigen Papillen bedingt den grauen Farbton der Zungenoberfläche. Manche Infektionskrankheiten verlangsamen die Abstoßung der Hornzellen, so daß sich der Zunge eine weißlich graue Schicht aus Hornschuppen und Bakterien auflagert ("belegte Zunge").
